Conventional and microwave-assisted specimen preparation methods for cultured cells and biomedical and plant tissues, for the benefit of those who only require routine TEM images through ultramicrotomy sectioning and then positive staining.
Cryo-specimen preparation by high-pressure freezing and cryoultramicrotomy.
Negative staining and immunogold labeling techniques for samples prepared through conventional, cryosectioning, or high-pressure freezing methods. Some of these chapters are presented in correlative approaches using TEM with fluorescent or confocal microscopy. Quantitative aspects of immunogold labeling in resin embedded samples are also included.
TEM crystallography and cryo-TEM tomography for the study of membrane proteins, macromolecules, organelles, and cells. One chapter on the application of EELS to biomedical tissue is included.
